Automatic Quantification for Scratch Wound Healing Assay

ADCIS developed a very innovative software application to automatically quantify wound healing over time. In this application, the quantification is efficiently performed in a more accurate, reliable, and reproducible manner.
Wound Healing Study - Role of HA molecular
This application is applied to evaluate the role of HA molecular weight in the proliferative phase of wound healing (Hyaluronic Acid).
Protocol
Scratch wound assay: Monolayer is wounded by a manual scratch with a pipette tip. Solutions containing different HA at 0.2% in culture medium with 2.5% of FCS are distributed at Day 0 and cultures are kept at 37 Celsius degrees for 24 hours (Day 1). Cells are placed in a Petri dish to study their evolution in time.
Results
The goal of the study is to compare the healing wound over time in function of the HA molecular weight. A comparison between high, medium, and low molecular weights is automatically performed.

Measurement Analysis and conclusion

After 24 hours, medium molecular weight of Hyaluronan (MMW-HA) has highly increased cell proliferation compared to the control Petri dish that has a mono-layer cell population with no hyaluronic acid. Image processing helped to automatically extract the healing area and compute its surface area. This method is more robust and more accurate than a manual drawing, and also much faster.
